I need to calculate routes from a single start point to over 1000 different end points. I can import list of end points PostCodes [ZipCodes] from Excel. Is there a way to automate this process without having to key over 1000 end point codes to obtain route?

Yes, with programming.. the full routing functionality is exposed in MapPoint object model. I don't have a lot of experience with the routing methods, but look in the help file, it should be straightforward for a VB programmer (couple hours at most) to figure out.
