Here is what I want to do: Take a map of downtown Houston, TX and place points on various business locations to see how close they are to the Metro rail line. I was planning to buy a map and put tacks on it, but a friend said mappoint would be a lot easier. Will it do this for me?

Yes it will. You can also draw the Metro Line as a line to make it clearer.

You could also do this with the much cheaper Streets & Trips. MapPoint adds a lot of demographic data and business data mapping options. Also it can be extended with custom or third party extensions.
